Introduction

The cultivation of soybeans is a vital component of agriculture in Indonesia, with the country being one of the largest producers of soybeans in the world. However, the process of breeding high-quality soybean seeds can be a time-consuming and labor-intensive process. To address this challenge, researchers have been exploring the use of in vitro culture techniques to accelerate the breeding process. In vitro culture involves the growth of plant cells or tissues in a controlled laboratory environment, allowing for the rapid multiplication of desirable traits.

Research conducted by Arsad Taher Harahap on vitro culture in various soybean genotypes (glycine max L. merrill) aims to analyze the effect of Auxin and cytokinin growth regulators (ZPT) on vitro breeding in soybeans. This study took place in the network culture laboratory of the Faculty of Agriculture, University of North Sumatra, Medan, from November 2002 to February 2003 under the guidance of Ir. Hot Setiado, MS as chairman and Ir. Hasmawi Hasyim, MS as a member.

Research Methodology

In this study, factorial complete random designs were used consisting of two factors. The first factor is variety (V) which has 23 levels, and the second factor is the concentration of ZPT (2PT) consisting of 5 levels. Thus, the total treatment combination reached 115, which was repeated twice with each treatment using two explants. Overall, this study involved 460 explants.

Observed parameters include:

Contaminated percentage: The percentage of explants that became contaminated with bacteria or fungi.
Percentage of growing: The percentage of explants that grew successfully.
Shoot height (cm): The height of the shoots that grew from the explants.
Number of leaves (strands): The number of leaves that grew from the explants.
Root length (mm): The length of the roots that grew from the explants.
Root volume (mm): The volume of the roots that grew from the explants.
When callus appears (MST): The time it took for callus to appear on the explants.
Callus weight (g): The weight of the callus that grew on the explants.
Callus color: The color of the callus that grew on the explants.

Research Results and Discussion

The results showed that the treatment of varieties had a significant effect on several growth parameters, such as shoot height, number of leaves, root length, root volume, when callus appeared, and callus weight. Conversely, there is no significant difference in the percentage of contamination, the percentage of growing, and the amount of roots.

On the other hand, ZPT treatment also has a significant effect on shoot height, number of leaves, root length, root number, root volume, when callus appears, and callus weight. However, for contaminated percentage parameters and growing percentages, the effect is not significant.

The interaction between varieties and ZPT treatment shows that there is no significant effect on all observed parameters. This shows that although the two treatments have a fairly strong impact individually, both do not affect each other when applied together.

Analysis and Implications

The findings of this study have significant implications for the development of soy tissue culture techniques. The selection of appropriate varieties and regulation of ZPT concentration can be optimized to increase soybean growth in vitro. Especially for farmers and plant breeders, understanding the effect of varieties and ZPT will open up opportunities to produce high-quality soybean seeds with the desired growth character.

In addition, this research highlights the importance of further research to explore the combination of varieties and other ZPT treatment, as well as understanding the mechanisms that underlie plant growth in vitro culture. With these steps, we can increase efficiency and effectiveness in plant breeding, which in turn has a positive impact on soybean productivity in Indonesia.

In addition, the results of this study can also be used as a reference for further research in order to improve tissue culture techniques in other plants, enrich scientific literature, and support agricultural sustainability through innovative biotechnology technology.

In conclusion, this study has demonstrated the potential of in vitro culture techniques in accelerating the breeding process of soybeans. The findings of this study have significant implications for the development of soy tissue culture techniques and highlight the importance of further research in this area. With the increasing demand for high-quality soybean seeds, the use of in vitro culture techniques can play a crucial role in meeting this demand and improving soybean productivity in Indonesia.
