Introduction

Griseofulvin is a widely used antifungal medication that has been employed to treat various fungal infections, including those affecting the skin, scalp, and nails. In this study, we aimed to enhance the dissolution of Griseofulvin and compare the bioavailability of Griseofulvin tablets in the form of solid dispersion and nanoparticles.

Research Methodology

To achieve our objectives, we employed two distinct methods to prepare Griseofulvin in the form of solid dispersion and nanoparticles. The solid dispersion was synthesized using the smelting method, while the nanoparticles were produced through the ball milling method. Furthermore, we characterized the particles using various techniques, including diffraction patterns, particle morphology, and the interaction between Griseofulvin and the carrier matrix. The solid dispersion was prepared using PEG 6000 as a carrier with a ratio of 2:8.

We also developed three different tablet formulations using direct printing methods: conventional tablets, solid dispersion tablets, and nanoparticle tablets. The dissolution test was conducted for 90 minutes, and the bioavailability measurements were carried out after administering the tablets to male rabbits (n = 5). The Griseofulvin levels in the blood were determined through spectrophotometry at a wavelength of λ = 291 nm. The bioavailability parameters tested included AUC, CMACS, and TMax, with significance specified through One Way Anova analysis (p <0.05 considered significant).

Results and Discussion

The results of our study revealed that the average size of Griseofulvin particles in the nanoparticles was 17.3 nm. The dissolution test showed that the average cumulative percentage obtained from conventional Griseofulvin tablets, solid dispersion tablets, and nanoparticle tablets were 87.01%, 88.81%, and 92.46%, respectively. However, the results of the statistical analysis indicated that there were no significant differences in dissolution between the three dosage forms (p = 0.980).

In contrast, the bioavailability measurements showed more attractive results. The mean CMACS for each treatment were: conventional tablets (1.28 μg/ml), solid dispersion tablets (1.91 μg/ml), and nanoparticle tablets (5.20 μg/ml). The average TMax for conventional tablets and solid dispersion tablets was 3.3 hours, while nanoparticle tablets had a faster TMax of 2.6 hours. The AUCtotal value obtained for each preparation showed that conventional tablets had an AUC value of 74,745 μg/ml. Hours, solid dispersion tablets 97,313 μg/ml.
